This month my sister received a letter from NVC for her husband, and 3 children over age 21 for Visa Interview in September. All 3 children are above 21 years of age and in our case visa approval was delayed for 5 and half years so CSPA Rules apply and all 3 children visa fee is paid.

But now they receive another letter for Mumbai Consulate and it does not show 3 childrens name on the list and the interview date is same. What should I do?

It seems like NVC has already sent it to Mumbai, contact Mumbai Consulate immediately to resolve the issue. No sense contacting NVC at this point.

As long as NVC has all the five names, I shouldnt worry about it. Make sure all of them show up for interview!
